    Study on the expression and clinical significance of LncRNA-PVT1 in diabetic nephropathy

    • Objective To explore the changes of long non-coding RNA(LncRNA) -plasmacytoma variant translocation 1(PVT1) in diabetic kidney disease(DKD) and analyze its clinical significance.Methods A total of 76 patients with DKD(DKD group) and 81 patients with type 2 diabetes mellitus(T2DM group) were enrolled as the research objects between May 2019 and May 2021.A total of 79 healthy volunteers were enrolled as control group.The expression level of LncRNA-PVT1 in peripheral blood was detected by qPCR.The changes of LncRNA-PVT1, renal function and blood glucose in different groups were compared.The relationship between LncRNA-PVT1 and renal function indexes, blood glucose indexes was analyzed by Spearman correlation analysis.The predictive efficiency of LncRNA-PVT1 for DKD occurrence was analyzed by operator operating characteristic(ROC) curves.Results Compared with control group, body mass index(BMI), LncRNA-PVT1, hemoglobin A1c(HbA1c), low-density lipoprotein cholesterol(LDL-C), urinary β2-microglobulin(MG), urinary α1-MG, urinary albumin-creatinine ratio(ACR), blood urea nitrogen(BUN), serum creatinine(Scr) and cystatin C(Cys C) were increased in DKD group and T2DM group(P<0.05), while high-density lipoprotein cholesterol(HDL-C) was decreased(P<0.05).Compared with T2DM group, LncRNA-PVT1, HbA1c, LDL-C, urinary β2-MG, urinary α1-MG, ACR, BUN, Scr and Cys C were increased in DKD group(P<0.05), while HDL-C level was decreased(P<0.05).LncRNA-PVT1 expression was positively correlated with HbA1c, LDL-C, urinary β2-MG, urinary α1-MG, ACR, BUN, Scr and Cys C R levels(P<0.001), while negatively correlated with HDL-C level(P<0.001).The area under the curve(AUC) and cut-off value of LncRNA-PVT1 for predicting DKD occurrence were 0.907(95%CI:0.862-0.953, P<0.001) and 1.830, respectively.Conclusions The expression of LncRNA-PVT 1 increases in DKD, which is correlated with renal function and blood glucose level, and also has high predictive value for DKD occurrence.
