An experimental study renal cancer cell apoptosis induced by picrosideⅡthrough mitochondrial pathway

Abstract
Objective To study the effect of picroside Ⅱ on the apoptosis of renal carcinoma ACHN cells and its mechanism.Methods In vitro cultured renal carcinoma ACHN cells were divided into control group and different picroside Ⅱ concentration(1 μg/mL,10 μg/mL and 100 μg/mL)groups.Cell proliferation rate were determined by CCK-8 method,cell apoptosis were observed by Hoechst 33258 nuclear staining and Annexin V/PI.The expressions of Bax and Bcl-2 in renal carcinoma ACHN cells were detected by Western blot method.Results The results showed that different concentrations of picroside Ⅱ could inhibit the growth of renal carcinoma ACHN cells and induce apoptosis.With the increase of picroside Ⅱ concentration,the apoptotic rate of renal carcinoma ACHN cells increased in a dose-dependent manner.After picroside Ⅱ treatment,the protein expression of Bax increased significantly and Bcl-2 expression decreased significantly.Conclusions Picroside Ⅱ inhibits the proliferation of renal carcinoma ACHN cells and promote the apoptosis of renal carcinoma ACHN cells,by changing the expressions of the apoptosis-associated signaling molecules Bax and Bcl-2 in the mitochondrial apoptosis pathway.
