Expression of miRNA and TSP-1 mRNA in renal tubular epithelial cells

Abstract
Objective To study the expression of miR-18a, miR-19a, miR-194 and TSP-1 in renal tubular epithelial cells treated with high sugar and TGF-β1.Methods The mRNA expression levels of TSP-1, miR-18a, miR-19a and miR-194 in human renal tubular epithelial cells in low glucose group (final concentration of glucose:5 mmol/L), high glucose group (final concentration of glucose:30 mmol/L) and high glucose+TGF-β1 group were detected by RT-PCR.Results With the rise of blood glucose and prolongation of the incubation time, the expression level of TSP1 mRNA increased gradually, and miR-194 decreased gradually, but the expression levels of miR-18a and miR-19a had no significant change. The mRNA expression of TSP1 was significantly up-regulated, and that of miR-194 was significantly down-regulated in high glucose+TGF-β1 group as compared with high glucose group.Conclusions TGF-β1 positively regulates the expression of TSP1, and negatively regulates miR-194. We need to confirm whether TSP1 is a target gene of miR-194 in the future.
