    铜死亡在肾脏疾病中的潜在作用

    Potential roles of cuproptosis in kidney disease

    • 摘要: 铜稳态在肾脏疾病的发生和发展中具有重要意义。铜死亡已在包括肾细胞癌的肿瘤领域广为研究,但其在急性肾损伤和慢性肾脏病中鲜见报道。铜死亡相关基因脂酰转移酶1显示了对IgA肾病的预测价值;晚期慢性肾脏病患者血铜升高,在动物模型中证实铜螯合剂可以抑制肾脏纤维化。线粒体功能障碍是铜死亡的标志。肾小管细胞富含线粒体易受活性氧簇攻击,且高表达铜转运蛋白,在药物的摄取和转运中发挥重要作用。因此,本文旨在总结铜死亡在肾脏疾病中的研究进展并探寻铜死亡与急性肾损伤的潜在联系,以期为肾脏疾病的治疗提供新思路。

       

      Abstract: Copper homeostasis plays an important role in the occurrence and development of kidney diseases. Cuproptosis has been widely examined in the field of tumors including renal cancer. However, it is rarely reported for acute kidney injury and chronic kidney disease. Cuproptosis related gene lipoacyltransferase 1 has shown its predictive value for IgA nephropathy. Blood copper levels became elevated in patients with advanced chronic kidney disease. And copper-chelating agents could arrest renal fibrosis in animal models. Mitochondrial dysfunction is a marker of cuproptosis. Renal tubular cells are rich in mitochondria and susceptible to injuries of reactive oxygen species. A high expression of copper transporters plays an important role in drug uptake and transport. Summarizing the latest researches of cuproptosis in kidney diseases, this review focused upon a potential correlation between cuproptosis and AKI and provided novel therapeutics for kidney diseases.
