    LncRNA HOTAIR、VEGF在肾上腺恶性肿瘤组织中的表达及其预后相关性

    Expressions of LncRNA HOTAIR and VEGF in adrenal malignant tumors and their prognostic significance

    • 摘要: 目的 研究肾上腺恶性肿瘤患者长链非编码RNA(long non-coding RNA,LncRNA)人类同源盒基因转录反义间RNA(HOX transcript antisense intergenic RNA,HOXTAIR)、血管内皮生长因子(vascular endothelial growth factor,VEGF)表达水平,并分析其与患者预后的关系。方法 选取2014年1月至2017年1月于沧州市人民医院手术治疗切除的肾上腺标本167例患者。采用实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)法检测所有研究对象组织HOTAIR、VEGF mRNA表达水平。Pearson法分析肾上腺恶性肿瘤组织HOTAIR与VEGF的相关性。Kaplan-Meier法评估HOTAIR、VEGF mRNA表达水平与肾上腺恶性肿瘤患者生存情况关系。采用COX法分析影响肾上腺恶性肿瘤患者预后的因素。结果 与良性肾上腺组织相比,肾上腺恶性肿瘤组织HOTAIR、VEGF mRNA相对表达水平较高(P<0.05)。HOTAIR与VEGF呈正相关(r=0.739,P<0.05)。HOTAIR、VEGF mRNA与肾上腺肿瘤临床病理特征的关系显示HOTAIR、VEGF mRNA与有无远处转移相关(P<0.05),与年龄、性别、肿瘤直径、肿瘤单发或多发无关(P>0.05)。HOTAIR低表达肾上腺肿瘤患者平均生存时间为(32.09±0.92)个月,显著长于HOTAIR高表达肾上腺肿瘤患者(21.64±1.49)个月(P<0.05);VEGF mRNA低表达肾上腺肿瘤患者平均生存时间为(31.81±0.97)个月,显著长于VEGF mRNA高表达肾上腺肿瘤患者(22.37±1.47)个月(P<0.05)。多因素分析表明,远处转移、HOTAIR高表达、VEGF高表达是影响肾上腺肿瘤患者不良预后的独立危险因素。结论 LncRNA HOTAIR、VEGF在肾上腺恶性肿瘤组织表达上调,可能是肾上腺肿瘤患者不良预后的生物标志物。

       

      Abstract: Objective To explore the expression levels of long non-coding RNAHOX transcript antisense intergenic RNA(LncRNAHOTAIR)and vascular endothelial growth factor(VEGF)in patients with adrenal malignant tumor,and analyze their prognostic relationship.Methods From January 2014 to January 2017,167 cases of adrenal gland specimens were selected.The expression levels of HOTAIR and VEGF mRNA were detected by real-time fluorescent quantitative PCR(qRT-PCR).Pearson’s method was employed for analyzing the correlation between HOTAIR and VEGF.And Kaplan-Meier method was applied for evaluatimg the relationship between HOTAIR,VEGF mRNA expression levels and survival of patients with adrenal malignant tumor.In addition,COX method was utilized for analyzing the prognostic factors of patients with adrenal malignant tumor.Results Compared with those in benign adrenal tissues,the relative expression levels of HOTAIR and VEGF mRNA were higher in malignant adrenal tumors(P<0.05).HOTAIR was positively correlated with VEGF(r=0.739,P<0.05).The relationship between HOTAIR,VEGF mRNA and clinicopathological features of adrenal tumors showed that HOTAIR and VEGF mRNA were correlated with distant metastasis(P<0.05),but not with age,gender,tumor diameter,single or multiple tumors(P>0.05).The mean survival time of adrenal tumor patients with low HOTAIR expression was(32.09±0.92)months.It was significantly longer than(21.64±1.49)months of adrenal tumor patients with high HOTAIR expression(P<0.05);The mean survival time of adrenal tumor patients with low VEGF mRNA expression was(31.81±0.97)months and it was significantly longer than(22.37±1.47)months of adrenal tumor patients with high VEGF mRNA expression(P<0.05).Multivariate analysis indicated that distant metastasis,high expressions of HOTAIR and VEGF were independent risk factors for poor prognosis of patients with adrenal tumors.Conclusions The expressions of LncRNA HOTAIR and VEGF are up-regulated in adrenal malignant tumors.Both may be biomarkers of poor prognosis in patients with adrenal tumors.
