    二甲双胍通过调节组蛋白乙酰化抑制肾癌细胞786-O迁移和侵袭

    Metformin inhibits migration and invasion of renal carcinoma cell line 786-O by regulating acetylation of histone

    • 摘要: 目的 研究二甲双胍抑制肾癌细胞迁移和侵袭的机制。方法 采用transwell迁移和matrigel侵袭实验检测10 mM和20 mM二甲双胍对786-O细胞迁移和侵袭的影响;Western blot实验检测二甲双胍对E-cadherin、vimentin、MMP-2、MMP-9以及组蛋白H3K9、H4K12和H4K16乙酰化水平的改变;组蛋白乙酰转移酶抑制剂C646与二甲双胍联用,通过transwell实验观察二甲双胍调控组蛋白乙酰化对786-O细胞迁移的影响。结果 10 mM和20 mM二甲双胍均能显著抑制786-O细胞迁移和侵袭(P<0.05);二甲双胍上调E-cadherin,下调vimentin、MMP-2、MMP-9的表达(P<0.05),同时显著增加组蛋白H3K9、H4K12和H4K16的乙酰化水平(P<0.05);C646和二甲双胍联用能显著阻止二甲双胍引起的迁移抑制作用(P<0.05)。结论 二甲双胍通过上调组蛋白乙酰化水平抑制肾癌细胞786-O的迁移和侵袭。

       

      Abstract: Objective To investigate the mechanism of metformin inhibition of migration and invasion of renal carcinoma cell line 786-O.Methods Transwell migration and Matrigel invasion assays were used to examine the role of metformin at 10 mM and 20 mM on migration and invasion of the cell line 786-O.Expression of E-cadherin,vimentin,MMP-2 and MMP-9 as well as acetylation of histone H3K9,H4K12 and H4K16 were detected by western blotting after metformin treatment.When combining the histone acetyltransferase inhibitor C646 with metformin,effect of metformin regulation of histone acetylation on migration of the cell line 786-O was observed through Transwell assay.Results Both 10 mM and 20 mM metformin remarkably inhibited migration and invasion of 786-O cells (P<0.05).The expression of E-cadherin was upregulated while expression of vimentin,MMP-2 and MMP-9 was downregulated after exposure to metformin (P<0.05).What's more,the acetylation levels of histone H3K9、H4K12 and H4K16 were significantly upregulated by metformin treatment(P<0.05).Combination of metformin with C646 could blocked inhibition of migration resulting from metformin treatment alone (P<0.05).Conclusions Metformin inhibits migration and invasion of renal carcinoma cell line 786-O through upregulation of histone acetylation.
