    Relationship between Wnt/β-catenin signaling pathway and lupus nephritis

    • 摘要: 目的 探讨Wnt/β-catenin信号通路中的分子在狼疮肾炎(LN)患者中的表达。方法 选取2015年5月至2016年4月在兰州大学第二医院就诊的LN患者18例作为LN组,另选取年龄、性别相匹配的健康体检者19名作为对照组。给予LN组激素和(或)激素联合免疫抑制剂治疗,对照组无特殊治疗。LN组患者经过6个月的激素及免疫抑制剂的治疗后,其中治疗有效8例,治疗无效10例。ELISA法检测所有研究对象血清中Wnt1、β-catenin、DKK1的表达。实时荧光定量PCR检测外周血单个核细胞中Wnt1、β-catenin、DKK1 mRNA的表达。结果 LN组患者与对照组Wnt1、β-catenin、DKK1及血清mRNA表达无统计学差异;LN组治疗无效患者血清中β-catenin表达高于治疗有效患者(P<0.05)。结论 Wnt/β-catenin信号通路的分子有望成为监测LN治疗无效的指标。

       

      Abstract: Objective To evaluate the Wnt/βcatenin signaling biomarkers in lupus nephritis (LN) patients. Methods Eighteen patients with lupus nephritis in Lanzhou University Second Hospital from 2015 to 2016 were included, and 19 healthy people served as normal control group. The expression of Wnt1, β-cateninand DKK1 mRNA was detected by real-time polymerase chain reaction, and their plasma concentrations were determined by ELISA. Results There was no significant difference in plasma levels and mRNA expression of Wnt1, β-catenin and DKK-1 between LN group and normal control group.Theserum level of β-catenin was significantly higher in no responsive LN patients than in responsive ones (P<0.05).Conclusions Biomarkers in Wnt/β-catenin signaling pathway may be valuable for identification of responses for LN therapy.
